Inhibition of mast cell degranulation by saponins from Gleditsia sinensis--structure-activity relationships.

摘要: The objective of the present study was to investigate effects saponins from anomalous fruits Gleditsia sinensis on mast cell degranulation triggered by compound 48/80 and determine possible structure-activity relationships. Among eleven tested, four effectively inhibited beta-hexosaminidase release rat peritoneal cells. Studies relationships indicated that with an aglycone echinocystic acid instead oleanolic were more effective. A sugar chain at C-3 essential for inhibitory effects, a single most length structures oligosaccharide C-28 also critical activity compounds, introduction monoterpene units substantially decreased activity. To gain insight into mechanisms responsible preventing degranulation, intracellular cAMP examined. After preincubation cells different times, active significantly increased content. These findings suggested might be constituents G. antiallergic activities, they prevented probably elevating levels.
