摘要: The effects of the phospholipase C (PLC) inhibitor U73122 on intracellular calcium levels ([Ca2+]i) were studied in MDCK cells. elevated [Ca2+]i dose-dependently. Ca2+ influx contributed to 75% 20 microM U73122-induced signals. pretreatment abolished transients evoked by ATP and bradykinin, suggesting that inhibited PLC. signals among individual cells varied considerably. internal source for response was endoplasmic reticulum (ER) since thapsigargin. depletion ER store triggered a La3+-sensitive capacitative entry. Independently release Ca2 entry, directly through La3+-insensitive pathway. augmented carbonylcyanide m-chlorophynylhydrozone (CCCP), but not Na+ removal, implicating mitochondria significantly buffering signal, efflux via Na+/Ca2+ exchange insignificant.
