摘要: Lipoprotein receptor-related protein 6 (LRP6) plays a critical role in skeletal development and homeostasis adults. However, the of LRP6 mesenchymal stem cells (MSCs), that give rise to osteoblastic lineage, is unknown. In this study, we generated mice lacking expression specifically nestin(+) MSCs by crossing nestin-Cre with LRP6(flox) investigated functional changes bone marrow alterations. Mice deletion demonstrated reductions body weight length at 1 3 months age. Bone architecture measured microCT (µCT) showed significant reduction mass both trabecular cortical homozygous heterozygous mutant mice. A dramatic numbers osteoblasts but much less osteoclasts was observed Osterix(+) osteoprogenitors osteocalcin(+) significantly reduced secondary spongiosa area, only moderately decreased primary area from colony forming, cell viability proliferation. Thus, essential for their survival proliferation, therefore, key positive regulator formation during growth remodeling.
