Characterization of the human CD4+ T-cell repertoire specific for major histocompatibility class I-restricted antigens

摘要: While CD4+ T lymphocytes usually recognize antigens in the context of major histocompatibility (MHC) class II alleles, occurrence MHC class-I restricted cells has been reported sporadically. Taking advantage a highly sensitive tetramer-based enrichment approach allowing detection and isolation scarce Ag-specific cells, we performed systematic comparative analysis HLA-A*0201-restricted CD8+ T-cell lines directed against several immunodominant viral or tumoral antigens. every peptide-MHC I complexes tested were detected all donors. These yielded strong cytotoxic helper 1 cytokine responses when incubated with HLA-A2+ target carrying relevant epitopes. HLA-A2-restricted seldom expanded immune donors, suggesting that they are not engaged vivo corresponding complexes. However, these expressed TCR very high affinity following ex stimulation by tumor cells. Therefore, describe versatile efficient strategy for generation could be used adoptive transfer- gene transfer-based immunotherapies.
