Resveratrol alleviates endotoxemia-associated adrenal insufficiency by suppressing oxidative/nitrative stress

摘要: We have recently demonstrated that endotoxin causes oxidative stress and overproduction of nitric oxide in adrenal glands, thereby leading to adrenocortical insufficiency. The aim this study is investigate the effects resveratrol, a natural plant polyphenol with anti-oxidant anti-nitrative properties, on endotoxemia-associated Resveratrol was administered immediately before injection lipopolysaccharide (LPS). Twenty four hours later, adrenocorticotropic hormone (ACTH) stimulation tests been performed measure plasma corticosterone level gland tissues were collected for histopathologic examination, determination malondialdehyde (MDA), total antioxidant capacity (T-AOC), superoxide dismutase (SOD) activity, catalase (CAT) inducible synthase (iNOS) expression, (NO) peroxynitrite production. Treatment resveratrol significantly inhibited endotoxemia-induced iNOS NO production, formation also attenuated LPS-induced gland, as evidenced by decrease pro-oxidant biomarker increases biomarkers (T-AOC, CAT SOD activity). H&E staining administration LPS resulted increased into gland. H&E-stained sections glands signs leukocyte infiltration hemorrhage during endotoxemia, which improved treatment. In addition, reversed downregulation ACTH receptor silent information regulator 1 (SIRT1) well hyporesponsiveness ACTH. exerts protective against insufficiency suppressing oxidative/nitrative stress. These findings support potential possible pharmacological agent improve resulting from damage.
