摘要: α-Fetoprotein (AFP) is a fetal serum protein abundant in liver whose expression downregulated adult liver. The promoter–proximal region of the AFP gene contains two binding sites for CCAAT/enhancer-binding α (C/EBPα), hepatocyte nuclear factor-1α (HNF-1α) and one factor-1 (NF-1). Luciferase reporter assays showed that combination C/EBPα, HNF-1α, NF-1 coactivator p300 gave maximal activity but BRG1 BRM, ATPase subunit chromatin remodeling complex SWI/SNF, repressed transactivation by these transcription factors. Deletion analyses C/EBPα suggested recruited SWI/SNF caused repression. This repression may also play important roles downregulation hepatocytes.
