Cyclopiazonic acid induced p53-dependent apoptosis in the testis of mice: Another male related risk factor of infertility.

摘要: Cyclopiazonic acid (CPA) is an indole tetrameric mycotoxin. This study carried out to investigate the potential effects of CPA on male reproductive system. In current study, 40 adult mice were divided into five groups (n = 8). The control group did not expose CPA, while animals in vehicle-received group; received CPA-solvent (0.05% dimethyl sulfoxide) and third, fourth, fifth 0.03, 0.06, 0.12 mg/kg, body weight, respectively for 28 days. Morphometric morphological deviations, spermatogenesis indices, malondialdehyde (MDA) content, total thiol molecules (TTM) concentration, antioxidant capacity (TAC), protein carbonylation rate (CO), nitric oxide (NO) concentration examined. expression changes apoptotic genes (P53, Bcl-2, Caspase III) at mRNA level also evaluated by qPCR technique. Reduction Leydig Sertoli cells population, diameter seminiferous tubules, parameters was significant only that highest dose CPA. An increase MDA, NO, CO testicular tissue reduction TAC TTM observed CPA-exposed groups. Significant up-regulation (p < .05) P53 III down-regulation Bcl-2 gene found CPA-received These results are indicating detrimental testicles, which may attribute CPA-induced oxidative stress apoptosis.
