摘要: Abstract A mutant of canine parvovirus (CPV) was derived after culture a highly passaged isolate CPV in the NLFK feline cell line. The virus (CPV-102/10) differed from parental strain its antigenic type when tested with panel neutralizing monoclonal antibodies, and relative ability to replicate dog line or dogs. formed little replicative form DNA cells. Five single recombinants two double between CPV-102/10 wild were constructed by digesting purified viral restriction enzymes, ligating separated ends different strains, examining viruses isolated transfecting into cultures. Analysis recombinant showed that mutation(s) determining both host range differences mapped 64 73 map units genome, within capsid protein gene. Sequencing region revealed adjacent amino acids, resulting nonconservative predicted acid sequences viruses. These results show infect dogs their cultured cells is determined, at least part, conformation surface capsid.
