摘要: This study was designed to test if treating chicken sperm with i) the cyclodextrins 2-hydroxypropyl-β-cyclodextrin (HBCD) and methyl-β-cyclodextrin (MBCD) alone improve fresh, liquid-stored cryopreserved semen quality, or ii) cholesterol-loaded (CLCs): loaded cholesterol (HCLC) (MCLC) enhance quality for application assisted reproductive technologies. Three consecutive experiments were performed different concentrations of additives: Exp. 1: 1, 2, 4 mg HBCD MBCD in fresh liquid stored semen; 2: HCLC MCLC 3: 2 mg HBCD, HCLC, 1 mg MBCD, post-thaw storage semen. Sperm motility parameters assessed by CASA system comprehensive characteristics evaluated flow cytometry. Supplementation resulted lowest functional spermatozoa 24 h at 5 °C. After cryopreservation, CLCs showed significantly lower progressive velocity movement, exhibited percentage cells intact plasma membranes acrosomes, mitochondrial activity without apoptosis. These results indicated that did not viability after cryopreservation. However, treated higher proportion motile (28%; P < 0.01) together high potential (25%; P < 0.05) compared control (18%; 21%, respectively) 3 h storage. CLC, especially methyl-β-cyclodextrin, appeared be detrimental spermatozoa, causing apoptosis, impairment potential, damaged acrosomes membranes.
