Individual and combined effects of MDM2 SNP309 and TP53 Arg72Pro on breast cancer risk: an updated meta-analysis

摘要: The tumor suppressor gene TP53 and its negative regulator murine double minute 2 are involved in multiple cellular pathways. Two potentially functional single nucleotide polymorphisms (SNPs) MDM2 SNP309 R72P have been extensively investigated to be associated with breast cancer risk. However, the original studies as well subsequent meta-analysis, yielded contradictory results for individual effect of two SNPs on risk, plus that conflicting also existed combined effects This meta-analysis aimed clarify these genes We performed a publications total 9,563 cases 9,468 controls concerning polymorphism 19,748 19,962 R72P. Odds ratios (ORs) 95 % confidence intervals (CIs) were used assess strength association. In overall individuals SNP309TG genotype borderline higher risk than those TT (OR = 1.11, CI: 1.00–1.24, P heterogeneity 0.007), whereas CC or GC had no stratified analyses, significant association between observed Asian, but null found even various subgroups. Moreover, analysis should treated caution, suggested investigations warranted explore molecular mechanism TP53-MDM2 circuit genes.
