摘要: Metastasis-associsated in colon cancer 1 (MACC1), a newly identified oncogene, promotes tumor cell proliferation and invasion. In the present study, expression of MACC1, hepatocyte growth factor (HGF) its receptor, MET proto-oncogene (c-Met), was investigated human gastric tissues adjacent normal by immunohistochemistry. The association between levels proteins clinicopathological parameters tumors were statistically analyzed. Furthermore, lentiviral particles expressing MACC1 used to infect hepatic satellite (HSC) line LX2. α-smooth muscle actin (SMA), HGF, matrix metallopeptidase (MMP)-2 MMP-9 HSCs examined western blotting reverse transcription-quantitative polymerase chain reaction. Transwell assays measure effect MACC1-infected or non-infected on migration invasion abilities MKN45 MKN74 carcinoma cells vitro. results demonstrated that positive protein HGF c-Met significantly higher compared with tissues. Positive had no correlation sex, age, location peritoneal metastasis patients, but correlated size, depth invasion, lymph node metastasis, TNM stage, histological differentiation, overall (5 years) disease-free survival years). each be positively other Western confirmed overexpressed MACC1-overexpressing lentivirus-infected HSCs. Overexpression increased MMP-2, α-SMA Results from indicated an increase number migrating towards HSCs, control These findings suggested may regulate MMP-2 thus promote cells. might cooperatively participate malignant progression cancer. conclusion, serve as useful molecular target for diagnosis, treatment prognosis
