Melanoma Cells Selected for Adhesion to Laminin Peptides Have Different Malignant Properties

摘要: Laminin is an important promoter of cell-matrix interactions. A number active laminin domains have been defined by use synthetic peptides. The Tyr-Ile-Gly-Ser-Arg (YIGSR) sequence on the B1 chain in can decrease tumor growth and metastasis, whereas another containing Ser-Ile-Lys-Val-Ala-Val (SIKVAV) increase metastasis. Here, we selected B16-F10 melanoma cells adherence or nonadherence to either YIGSR- SIKVAV-coated dishes established 3 variants: YIGSR-adherent (Y+), YIGSR-nonadherent (Y-), SIKVAV-nonadherent (S-). SIKVAV-adherent were not because most F10 attached dish. These cell lines proliferated at same rate as parent equally laminin, collagen IV, fibronectin. Y+ produced rapidly growing tumors after s.c. injection twice many lung colonies parental i.v. injection. In contrast, Y- more slowly one-third relative S- yielded similar numbers but smaller than data suggest that interactions with YIGSR site are probably for both colony formation a target organ (lung) subsequent growth, while SIKVAV-containing may be growth.
