摘要: Hepatocyte transplantation is considered to be a promising therapy for patients with liver diseases. Induced pluripotent stem cells (iPSCs) provide an unlimited source the generation of functional hepatocytes. In this study, we generated iPSCs from porcine ear fibroblasts (PEFs) by overexpressing Sox2, Klf4, Oct4, and c-Myc (SKOM), developed novel strategy efficient differentiation hepatocyte-like following processes early development. The differentiated displayed phenotypes hepatocytes, exhibited classic hepatocyte-associated bio-functions, such as LDL uptake, glycogen storage urea secretion, well possessed metabolic activities cytochrome P-450 (CYP) 3A 2C. Furthermore, compared hepatocyte efficacy our protocol another published method, results demonstrated that could significantly improve morphological iPSCs. conclusion, study establishes method in vitro hepatocytes iPSCs, which represent cell preclinical testing cell-based therapeutics failure pharmacological applications.
