Mechanisms of drug resistance in colon cancer and its therapeutic strategies

摘要: Drug resistance develops in nearly all patients with colon cancer, leading to a decrease the therapeutic efficacies of anticancer agents. This review provides an up-to-date summary on over-expression ATP-binding cassette (ABC) transporters and evasion apoptosis, two representatives transport-based non-transport-based mechanisms drug resistance, as well their strategies. Different ABC were found be up-regulated which can facilitate efflux drugs out cancer cells effects. Inhibition by suppressing protein expressions or co-administration modulators has been proven effective approach sensitize drug-resistant vitro. On other hand, apoptosis observed cancers also results agents, especially inducers. Restoration apoptotic signals BH3 mimetics epidermal growth factor receptor inhibitors inhibition cell alternative death pathways, such autophagy, are means treat resistant types. Given that different among may change even single patient at stages, personalized specific combination therapy is proposed more safer for reversal clinics.
