摘要: A DNA vaccine, using a pCl-neo plasmid encoding the glycoprotein gene of Mexican isolate rabies virus, was developed to induce long-lasting protective immunity against virus in dogs. The aim this work evaluate intranasal (IN) vaccination route mice and Mice dogs were immunized via intramuscular (IM) IN routes. received 50 microg booster on day 30, same doses routes, 90 they challenged. Dogs 100 with 180, immune responses studied 210. Virus-neutralizing antibodies detected blood sera (up 0.5 IU) animals when submitted booster, levels neutralizing increased. Animals vaccinated IM presented higher antibody titres than those IN. Control groups lacked anti-rabies antibodies. On 90, From these, % mice, an 80 survived challenge. No from control group survived. revealed significant virus-neutralizing 30 and, after 210 titre 1.8 IU. main advantage vaccines over traditional live ones is that there no contamination viruses could be disseminated environment reproduced susceptible animals. This study demonstrated pGQH succesful administrated route. seems attractive due its easy application mucosal protection. form also advantageous domestic animal campaigns, for it less stressful parenteral (no painful shots).
