摘要: We examine the effects of aberrations induced by a refrac- tive index mismatch on signal level and resolution single- photon (12p) two-photon (22p), conventional confocal scanning microscopes. In particular, we consider in- troduced an interface between oil/glass water. Resolution is defined in terms enclosed fluorescence, rather than full-width half- maximum, revealing more useful information for heavily aberrated point spread functions (PSFs). It shown that, at large focusing depths, 22p 12p mi- croscopes are almost identical. The benefits aberration correction examined removing Zernike modes. With correction, best found An approximation based upon geometrical optics also introduced which shows that axial ab- errated PSFs roughly proportional to depth. © 2001 Society
