Protein kinase D1 deficiency promotes differentiation in epidermal keratinocytes

摘要: Summary Background Protein kinase D (PKD or PKD1) is a serine/threonine protein that has been shown to play role in variety of cellular processes; however, the function PKD1 skin not fully investigated. The balance between proliferation and differentiation processes predominant cells epidermis, keratinocytes, essential for normal function. Objective To investigate effect deficiency on epidermal keratinocytes. Methods We utilized floxed mouse model such infecting keratinocytes derived from these mice with an adenovirus expressing Cre-recombinase allowed us determine gene loss vitro . Proliferation were monitored using qRT-PCR, Western blot, transglutaminase activity assays, [ 3 H]thymidine incorporation into DNA cell cycle analysis. Results A significant decrease mRNA levels was achieved adenoviral Cre-recombinase-infected cells. Deficiency resulted increases expression various markers as loricrin, involucrin, keratin 10 either basally and/or upon stimulation differentiation. PKD1-deficient also showed increase activity, indicating anti-differentiative PKD1. Furthermore, exhibited decreased proliferation. However, had no stem marker expression. Conclusions Cre-recombinase-mediated knockdown represents additional approach demonstrating anti-differentiative, pro-proliferative signal
