Isolation and characterization of an alphoid centromeric repeat family from the human Y chromosome.

摘要: A collection of human Y-derived cosmid clones was screened with a plasmid insert containing member the X chromosome alphoid repeat family, DXZ1. Two positive cosmids were isolated and repeats they contained investigated by Southern blotting, in situ hybridization sequence analysis. On to genomic DNAs, expected cross-hybridization characteristic all sequences seen and, addition, 5500 base EcoRI fragment found be Y-specific repeat. Dosage experiments demonstrated that there are about 100 copies this on Y chromosome. Studies utilizing DNA from human-mouse hybrids only portions hybridizations spreads centromeric localization Cross-hybridization autosomes 13, 14 15 also seen; however, these chromosomes lacked detectable arrangement. Sequence analysis DXZ1 70% homology each other consensus sequence. The not gorilla, orangutan or chimpanzee male DNA.
