The Number of Direct Repeats in hagA Is Variable among Porphyromonas gingivalis Strains

摘要: The coding sequence for the surface protein hemagglutinin A (HagA) of Porphyromonas gingivalis 381 has previously been shown to contain four direct 1.35-kb repeats, designated repHA. This study was performed determine if number repHA units in hagA is consistently 4 or allelic polymorphism exists among strains and/or upon multiple passage P. gingivalis. To this end, primers which were homologous regions directly 5' and 3' repeat domain synthesized. PCR conditions allowed amplification 8.4-kb region between established. Genomic DNA templates from 13 other 9 fresh clinical isolates patients analyzed under same as used above. Analysis these products demonstrated that tested had different numbers (two four) respective genes. 8.4, 7.0, 5.7 kb represent four, three, two respectively. One strain each group (381, repeats; W83, three AJW4, repeats) also repeats remained invariant passaging onto solid medium. No variability within a detected after 18 passages. chosen further testing mouse abscess model vivo growth would select deletions duplications repeated sequences. Five days infection, no change cells recovered either nonimmunized preimmunized mice. data indicates an interstrain hence size HagA gingivalis, but unlike some antigens pathogenic species, remains relatively stable given here.
