Changes in matrix metalloproteinases and their endogenous inhibitors during tumor progression in the uterine cervix

摘要: To study the role of matrix metalloproteinases (MMPs) and tissue inhibitors (TIMPs) in cervical tumorigenesis, we analyzed 70 specimens that included 15 low-grade squamous intraepithelial lesions (SILs), 20 high-grade SILs, 25 cell carcinomas (SCCs) 10 normal tissue. The gelatinolytic activity MMP-9 MMP-2 was determined by zymographic analysis. expression TIMP-1 TIMP-2 immunohistochemistry. All samples had 72/66 kDa gelatinase activity; 92 kDa detected only SILs SCCs. Immunohistochemical analysis showed weak positivity for epithelium SILs. However, SCCs intense cellular stromal reactivity MMP-9. For TIMP-2, immunostaining, >50% positivity, 95% reactivity. Increase relative these gelatinases enhanced immunostaining MMPs TIMPs with tumor progression suggest they may play a crucial cancer progression. A significant association between stage lesion (P<0.01) found. studies indicate be basal origin tissue, although mechanism their upregulation is not clearly understood.
