MicroRNA-19b-3p promotes cell proliferation and osteogenic differentiation of BMSCs by interacting with lncRNA H19

摘要: To investigated the role of miR-19b-3p in regulating bone marrow mesenchymal stem cell (BMSC) proliferation and osteoblast differentiation. The expression lncRNA H19 were measured postmenopausal osteoporosis patients BMP-22 induced BMSCs using qRT-PCR. MiR-19b-3p mimic or inhibitor was transfected into BMP-2 BMSCs. Cell by BrdU method. Protein RUNX2 COL1A1 western blot. PcDNA3.1-lncRNA with without significantly up-regulated overexpression dramatically elevated, while inhibition decreased Additionally, protein levels COL1A1, as well ALP activity promoted transfection inhibited transfection. LncRNA obviously down-regulated patients. differentiation down-regulating miR-19b-3p. Moreover, inhibited, elvated 17β-estradiol (E2) treated a dose-dependent manner. These data first to reveal critical H19/miR-19b-3p osteoporosis, provided new therapeutic target for OP.
