Early Effect of Amyloid β-Peptide on Hippocampal and Serum Metabolism in Rats Studied by an Integrated Method of NMR-Based Metabolomics and ANOVA-Simultaneous Component Analysis.

摘要: Amyloid β (Aβ) deposition has been implicated in the pathogenesis of Alzheimer’s disease. However, early effect Aβ on metabolism remains unclear. In present study, thus, we explored metabolic changes hippocampus and serum during first 2 weeks Aβ25–35 injection rats by using an integrated method NMR-based metabolomics ANOVA-simultaneous component analysis (ASCA). Our results show that injection, time, their interaction had statistically significant effects metabolome. Furthermore, identified key metabolites mainly contributed to these effects. After from 1 weeks, levels lactate, N-acetylaspartate, creatine, taurine were decreased rat hippocampus, while increase lactate decreases LDL/VLDL glucose observed serum. Therefore, suggest reduction energy lipid as well anaerobic glycolysis may occur at stage deposition.
