摘要: Evodiamine has been reported to exhibit anti-inflammatory and anti-nociceptive effects, but the underlying mechanisms remain be defined. P2X4 receptor (P2X4R) is a subtype of ATP receptors plays important roles in pain, inflammatory immune responses. We aimed investigate whether evodiamine beneficial effects on endothelial injury mediated by chronic high glucose condition. found that culturing human umbilical vein cells (HUVECs) with significantly increased expression HUVECs, cytosolic Ca(2+) concentrations intracellular reactive oxygen species (ROS) while decreasing nitric oxide (NO); these could reversed evodiamine. High also pro-inflammatory activators (NF-κB) TNFR-ɑ, which was accompanied elevation P2X4R levels. able down-regulate elevated NF-κB, ROS, up-regulate decreased NO. Thus may exert anti-inflammation activity high-glucose challenge HUVEC via suppressing signaling pathway, exhibiting ability protect HUVECs from glucotoxicity.
