miR-144 may regulate the proliferation, migration and invasion of trophoblastic cells through targeting PTEN in preeclampsia

摘要: Previous studies indicated that microRNAs (miRNAs) were aberrantly expressed in the placentas of patients with Preeclampsia (PE); however, underlying mechanism still requires further investigation. The aim this study is to investigate roles miR-144 preeclampsia and related mechanism. expression PTEN 30 PE normal was compared; next, HTR8/SVneo cells transfected mimics inhibitors cultured for 48h, proliferation apoptosis, cell migration invasion examined; furthermore, PTEN, Caspase-3 Bcl-2 dual luciferase reporter assay has been performed confirm a direct target miR-144; finally, HTR-8/SVneo either overexpression plasmid or RNAi determine whether knockdown can mimic effect We have observed significantly decreased markedly increased compared placentas; moreover, transfection trophoblastic induced significant increase proliferation, migration, invasion, decrease also affected cycles; on other hand, shown opposite effects; transient marked (P<0.01), while showed confirmed as results inhibitor mimics, respectively. In conclusion, down-regulated PE, may play important pathogenesis through targeting cells. These suggested potential become therapeutic treatment PE.
