The Effect of e-, i-, and n-Nitric Oxide Synthase Inhibition on Colonic Motility in Normal and Muscular Dystrophy (Mdx) Mice

摘要: To explore the origin of diarrhea or constipation in human Duchenne muscular dystrophy (DMD), effect inhibition e- , i-, and n-nitric oxide synthase (NOS) on motility proximal distal segments colon (mdx) control mice was studied. The frequency migrating motor complexes (MMC) higher than mdx (0.56 vs. 0.25 cpm) (0.7 cpm), there no difference when compared to segments. High concentrations NOS inhibitors, including 1,3-PBIT dihydrobromide (1,3-PBIT) spermine, inhibited MMC. dose spermine required inhibit MMC lower for colon. In presence tetrodotoxin, (1 mM) (5 reduced magnitude local, rhythmic contractions (LC) paced by interstitial cells Cajal (ICC), but (50 increased their magnitude. There LC between results suggest an high e-, n-NOS modulation ICC activity e-NOS, greater susceptibility colon, which is very likely because a deficit smooth muscle affecting pacemaker. A pacemaker may be DMD.
