摘要: Abstract A set of five carbohydrate-specific monoclonal antibodies (mAbs) were used to probe the ultrastructure walls soybean pathogen Phytophthora megasperma f.sp. glycinea , using a combination immunofluorescence and immuno-gold labelling techniques. Results with β-1,3 glucan-specific suggest that glucans are present throughout both germ tubes cysts, but more prevalent in outer portion. In addition, on surface hyphal walls, not closely associated other material, most likely protein, sterically hinders antibody binding except non-reducing terminal residues. An whose epitope involved β-1,4 glucosyl linkages bound predominantly inner portion wall. However, fluorescent this suggested exterior P. as well. Staining another indicates changes wall composition occur over 50–100 μm from tip, greater distance than previously supposed. The role antigens recognized by these mAbs plant-pathogen interaction is known, potential uses discussed.
