Distribution of apoptosis-mediating Fas antigen in human skin and effects of anti-Fas monoclonal antibody on human epidermal keratinocyte and squamous cell carcinoma cell lines

摘要: Fast antigen is a cell surface protein that mediates apoptosis. Using immunohistological, flow cytometry and electron microscopic analyses, we investigated the expression of Fas on various skin tissues, cultured SV40-transformed human epidermal keratinocyte line KJD squamous carcinoma HSC. The was widely distributed in components such as keratinocytes lower portion epidermis, dendritic cells, endothelial fibroblasts, apocrine glands, eccrine sweat sebaceous some normal melanocytes infiltrating lymphoid cells. It also strongly expressed lichenoid eruptions seen lupus erythematosus lichen planus, spongiotic or acanthotic epidermis chronic eczema, adult T-cell leukaemia/lymphoma (ATLL) atopic dermatitis. Its closely correlated with cells it neoplastic particularly ATLL fibroblasts dermatofibroma. However, not detected basal epithelioma malignant melanomas any junctional naevi. lines HSC antigen, crosslinking by an anti-Fas monoclonal antibody induced apoptosis these lines. These results indicate apoptosis-mediating may play important role turnover differentiation, immune regulation tumours, pathogenesis diseases.
