The RNA Methyltransferase Dnmt2 Is Required for Efficient Dicer-2-Dependent siRNA Pathway Activity in Drosophila

摘要: Transfer RNA (tRNA) fragmentation in response to stress conditions has been described many organisms. tRNA fragments have found association with small interfering (siRNA) components, but the biological role of these interactions remains unclear. We report here that methyltransferase Dnmt2 is essential for efficient Dicer-2 (Dcr-2) function Drosophila. Using (sRNA) sequencing, we confirmed limits extent during heat-shock response. tRNAs as well serve Dcr-2 substrates, and degrades tRNA-derived sequences, especially under conditions. RNAs are able inhibit activity on long double-stranded (dsRNAs). Consequently, heat-shocked mutant animals accumulate dsRNAs, produce fewer siRNAs, show misregulation siRNA pathway-dependent genes. These results reveal impact pathways implicate modifications regulation sRNA homeostasis
