摘要: Abstract The effect of abscisic acid (ABA), non-permeating osmoticum and desiccation treatment on storage protein synthesis during maturation somatic embryos Picea glauca (Moench) Voss. was examined. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) Western blot analysis demonstrated that some the major crystalloid matrix polypeptides were absent from maturing medium containing ABA low osmoticum. However, with polyethylene glycol-4000 (PEG) in combination resulted a spectrum resembling mature zygotic embryos. These proteins accumulated throughout an 8-week culture period, resulting threefold higher content than for same time absence PEG. structure distribution bodies cells these osmotically treated similar to Treatment 5·0-7·5% PEG prevented catabolism desiccation. optimal conditions embryo deposition 16 μM 7·5% 8 weeks followed by Analysis mRNAs vitro translation immunoprecipitation translated products showed mRNA profiles those similar. differences observed pattern embryos, therefore, indicates storage-protein response is part regulated at translational level. During regeneration plantlets rapidly utilized manner seedlings.
