Sensitivity of the detection of plum pox potyvirus by molecular assays

摘要: Recently, plum pox potyvirus (PPV) has been found in Basilicata, southern Italy, on plum, apricot and peach. In 1992-09, we started a large-scale survey to verify the effectiveness of diagnostic methods used during seasons when it is difficult reveal any presence virus. The assays were carried out by dot-blot hybridization stone-fruit cultivars normally planted this area. virus was found, hybridization, be present seven peach, four one plum. All plants 8–10 years old and, except for two cultivars, not displaying apparent symptoms spring 1992. Five peach intended use as primary sources propagation material, then selected further study, assayed 1992–11 ELISA RT-PCR. tests these consistently negative, while PCR positive. However gave positive results repeated 1993–05. These only suggest that material should tested techniques more sensitive than ELISA, but also question usefulness carrying phenological phases plant.
