Aspirin effect on early and late changes in acute lung injury in sheep.

摘要: There have been several studies that already explored the potential beneficial role of cyclo-oxygenase (CO) inhibitors on oleic acid (OA)-induced lung injury in different species. These report no significant effect CO inhibition, though thromboxane B2 (TxB2) was effectively blocked. However, recent indicate pre-treatment with aspirin (ASA) preserve gas exchange OA dogs. Aim our study has to evaluate effects low doses ASA exchange, hemodynamics, respiratory mechanics, prostanoids and histology OA-induced sheep. 0.09 ml/kg administered into right atrium 14 anaesthetized Six received a bolus (10 mg/kg i. v.) 30 min before OA, others saline as placebo. Pulmonary tissue pulmonary systemic system TxB2 6-keto-PGF1α, leukocytes platelets concentrations were measured throughout subsequent 3 h effected at end-experiment. The principal findings are: 1) reduces early vasoconstriction bronchoconstriction, parallelled by suppression generation; 2) late increase artery pressure airway resistance due is not inhibited ASA; 3) disturbance reduced ASA, whereas severe deterioration exaggerated 4) stability ratio (R) ≈1 ASA-group compared its fall ≈0.7 controls. Our suggest ASA: only effective treat very transient TxB2-induced resulting hydrostatic edema, it ineffective, even accentuates, major endothelial cell leading alveolar flooding unrelated TxB2; protective bronchospasm; biphasic behaviour benefit which lasts one hour then results worse exchange; an immediate, stabilizing, persisting R, contrasting hemodynamics PaO2.
