Dendritic Localization of the Translational Repressor Pumilio 2 and Its Contribution to Dendritic Stress Granules

摘要: Pumilio (Pum) protein acts as a translational inhibitor in several organisms including yeast, Drosophila , Xenopus and mammals. Two genes, Pum1 Pum2 have been identified mammals, but their function neurons has not identified. In this study, we found that mRNA is expressed during neuronal development the discrete particles both cell body dendritic compartment of fully polarized neurons. This finding indicates novel candidate dendritically localized ribonucleoparticles (RNPs). During metabolic stress, present stress granules (SGs), which are subsequently detected somatodendritic domain. It remains excluded from processing bodies under all conditions. When overexpressed fibroblasts, induces formation SGs also contain T-cell intracellular antigen 1 (TIA-1)-related protein, eukaryotic initiation factor 4E, poly(A)-binding TIA-1, other RNA-binding proteins Staufen1 Barentsz. induction dependent on domain glutamine-rich region N terminus Pum2. behaves similar manner TIA-1 prion two molecules with known roles aggregation. downregulation via RNA interference (RNAi) interferes stress.
