摘要: Using a combination of two-dimensional gel electrophoresis protein mapping and mass spectrometry analysis, we have established proteome reference maps embryogenic cell suspensions cowpea (Vigna unguiculata). The were generated from young primary leaves contained basically pro-embryogenic masses, which enabled us to dissect their composition while eliminating the complexity too many types. Over 550 proteins could reproducibly be resolved over pI range 3-10. A total 128 most abundant spots excised, digested in-gel with trypsin analyzed by tandem spectrometry. This identification 67 spots. Two identified as chitinase ribonuclease belonging family PR-4 PR-10 proteins, respectively. expression respective genes was confirmed RT-PCR pattern deposition in well developing seeds, roots, shoots flowers determined Western blot experiments, using synthetic antibodies raised against 14-amino acid peptide located close C-terminal region protein.
