Mass discrimination in matrix‐assisted laser desorption ionization time‐of‐flight mass spectrometry: A study using cross‐linked oligomeric complexes

摘要: The determination of the mass discrimination effect matrix-assisted laser desorption ionization spectrometry (MALDI MS) over a wide m/z range was made by determining relative amounts prepurified protein monomer and dimer species present in reconstituted mixtures. Protein complexes were covalently cross-linked with glutaraldehyde monomeric dimeric separated gel filtration chromatography. Individual recombined specific ratios analyzed both MALDI MS sodium dodecyl sulfate polyacrylamide electrophoresis. contributions peak areas doubly charged ions singlycharged generated during analyses to that singly-charged [M + H]+ measured. calculation for two different time-of-flight instruments showed loss factor ∼2.6 area as value doubled 30 000–145 000. An overall correction is given which includes effects associated sample preparation, desorption/ion formation, ion transmission detection.
