摘要: The multicellular layers(MCL) of human cancer cells is a three dimensional(3D) in vitro model for solid tumors which has been used primarily the assessment avascular penetration anti-cancer drugs. For drugs with problem, MCL represents good experimental that can provide clinically relevant data. Calcein-AM fluorescent dye demonstrates cellular vitality graded manner cell culture system. In present study, we evaluated use calcein-AM determination anti-proliferative activity agents DLD-1 colorectal cells. Optical sectioning confocal imaging was compromised photonic attenuation and barrier deep layers MCL. By contrast, measurement on cryo-sections provided feasible alternative. Cold pre-incubation did not enhance distribution to significant degree However, simultaneous drug appeared be possible treated conclusion, these data suggest drug-induced effect model.
