摘要: The Squash bug, Anasa tristis (De Geer), is a major piercing/sucking pest of cucurbits, causing extensive damage to plants and fruits, transmitting phytopathogens. No genomic resources facilitate field laboratory studies this were available; therefore the first de novo exome for destructive was assembled. RNA extracted from insects challenged with bacterial fungal immunoelicitors, fed on different cucurbit species, all life stages egg adult. All treatments replicates separately barcoded subsequent analyses, then pooled sequencing in single lane using Illumina HiSeq2000 platform. Over 211 million 100-base tags generated manner trimmed, filtered, cleaned, assembled into reference transcriptome Broad Institute Trinity assembly algorithm. annotated NCBIx NR, BLAST2GO, KEGG other databases. Of >130,000 total assemblies 37,327 identifying sequences candidate gene silencing targets immune, endocrine, reproductive, cuticle, physiological systems. Expression profiling adult immune response accomplished by aligning each biological replicate treatment controls A. transcriptome.
