摘要: Abstract The effects of cAMP on the expression ETA (ET-1 selective type) receptor mRNA and response to endothelin-1 (ET-1) were investigated in rat aortic smooth muscle cells primary culture, using reverse transcription polymerase chain reaction (RT-PCR) fura-2 microfluorometry cytosolic Ca2+ concentrations ([Ca2+]i). Incubation with 10 μM forskolin 3-isobutyl-1-methyl-xanthine (IBMX) induced an increase by 420 % control after 24 hours. ET-1 a biphasic [Ca2+]i, first transient second sustained phases. IBMX for hours increased [Ca2+]i 849 phase 286 phase, compared time-matched controls. From these results, we conclude that induces up-regulation increases responsiveness culture.
