摘要: We have previously shown that the human cytomegalovirus early UL4 promoter has upstream negative and positive cis-acting regulatory elements. In absence of region, element confers strong transcriptional activity. The contains a CCAAT box dyad symmetry binds cellular transcription factor NF-Y. effect is negated by viral IE2 protein (L. Huang, C.L. Malone, M.F. Stinski, J. Virol. 68:2108, 1994). investigated binding or to region. major was located between -168 -134 bp relative start site. This could be transferred heterologous promoter, it functioned in either orientation. Mutational analysis demonstrated core DNA sequence element, 5'-GTTTGGAATCGTT-3', required for repressor protein(s) protein. activity present both nonpermissive HeLa permissive fibroblast cells but more abundant cells. Binding correlates with repression from promoter. negation repressive effect.
