Direct transfer of Ku between DNA molecules with nonhomologous ends

摘要: Abstract The Ku protein is an essential for DNA double-strand-break repair by the pathway of nonhomologous end-joining (NHEJ). A previous study showed that bound to one molecule could transfer directly another without being released into solution first. Direct requires two molecules having homologous cohesive ends with a minimum four complementary bases. Results this reveal direct activity regulated NaCl and MgCl2. Increasing either cations can decrease required amount other. However, end-binding not affected changing concentration cations, indicating activities are independently. Most importantly, results also show from under condition 200 mM 5 mM ability between DNAs suggests align or juxtapose during NHEJ.
