Mechanism of oligonucleotide uptake by cells: involvement of specific receptors?

摘要: We have investigated the interaction of oligonucleotides and their alkylating derivatives with mammalian cells. In experiments L929 mouse fibroblast Krebs 2 ascites carcinoma cells, it was found that cellular uptake oligodeoxynucleotide is achieved by an endocytosis mechanism. Uptake considerably more efficient at low oligomer concentration (less than 1 microM), because this a significant percentage total pool absorbed on cell surface internalized absorptive process. Two modified proteins were detected in fibroblasts treated oligonucleotide derivatives. The binding oligomers to inhibited other oligodeoxynucleotides, single- double-stranded DNA, RNA. polyanions heparin chondroitin sulfates A B do not inhibit binding.
