摘要: Resistance to platinum-based chemotherapy is the major obstacle successful treatment of ovarian cancer. It evident that mitochondrial defects and dysfunctions oxidative phosphorylation energy production in cancer cells were directly related their resistance platinum drugs. Using 2-D DIGE, we compared proteins from two platinum-sensitive human cell lines (SKOV3 A2780) with four platinum-resistant sublines (SKOV3/CDDP, SKOV3/CBP, A2780/CDDP, A2780/CBP). Among 236 differentially expressed spots, five (ATP-α, PRDX3, PHB, ETF, ALDH) participate electron transport respiratory chain identified through mass spectrometry. All them are downregulated one or lines. Three PHB) validated by using western blot immunohistochemistry. There a significant decrease PHB tumor tissues patients who resistant chemotherapies. This first direct proteomic comparison between cells. These studies demonstrated DIGE-based analysis could be powerful tool investigate limited proteins, association expression indicates mitochondria may contribute
