摘要: Abstract Results of a number epidemiological and experimental studies indicate that polyphenols (e.g. resveratrol (RES), epicatechins etc.), antioxidant agents abundant micronutrients in our food could have strong anti-mitotic as well pro-apoptotic effects. In this study we raised the question whether roscovitine (ROSC), an inhibitor cyclin-dependent kinases (CDKs) with increased selectivity towards CDK2, be able to affect human leukemia HL-60 cells which p53 gene is inactivated ROSC-induced effects additionally modulated by compounds natural origin, especially e.g. RES. Exposure ROSC for 24 h inhibited their proliferation. Flow cytometric analyses revealed unlike MCF-7 cells, were arrested G1 upon treatment. Furthermore, also induced apoptosis cells. After treatment 40 μM frequency hypoploid representing undergoing reached approximately 50%. next step action RES alone or combination was examined. Interestingly, synergistic observed after combined sequential post-incubation 48 h presence Such resulted marked reduction S- G2/M-phase simultaneously cell population up 80% at fourfold lower dose. Further strongly activated caspase-3. These results clearly evidence potentiates apoptosis.
