Single-Dose and Steady-State Pharmacokinetics of Aminoglutethimide

摘要: The oral pharmacokinetics of aminoglutethimide were determined in 17 patients receiving the drug therapeutically. absorption after intake was almost complete as judged by recovery radio-labelled urine. plasma half-life markedly reduced (mean 43%) during multiple-dose administration compared with a single dose, but only moderate increase total clearance 26.9%) observed. This finding consistent significant reduction 29.2%) apparent volume distribution (Vd) occurring prolonged treatment. These alterations could also be demonstrated drug-free interval 96 hours not explained altered protein binding aminoglutethimide, evaluated equilibrium dialysis experiments.
