High-Frequency, In Vitro Reversible Switching of Candida lusitaniae Clinical Isolates from Amphotericin B Susceptibility to Resistance

摘要: Recent studies have revealed an increase in the incidence of serious infections caused by non-albicans Candida species. lusitaniae is special interest because its sporadic resistance to amphotericin B (AmB). The present vitro study demonstrated that, unlike other species, C. isolates frequently generated AmB-resistant lineages form previously susceptible colonies. Cells switching from a resistant colony phenotype were also detected after treatment with either UV light, heat shock, or exposure whole blood, all which increased frequency switching. In some lineages, cell switched phenotype, was stable; colonies composed primarily AmB-susceptible cells. Although and identical many aspects, their cellular morphologies dramatically different. Switching mechanisms that involve antifungals may impact on antifungal therapeutic strategies as well standardized susceptibility testing clinical yeast specimens.
