An H4K16 histone acetyltransferase mediates decondensation of the X chromosome in C. elegans males

摘要: In C. elegans, in order to equalize gene expression between the sexes and balance X autosomal expression, two steps are believed be required. First, an unknown mechanism is hypothesized upregulate chromosome both sexes. This balances males, but creates overexpression hermaphrodites. Therefore, restore balance, hermaphrodites downregulate twofold on chromosomes. While many studies have focused downregulation, of upregulation not known. To gain more insight into upregulation, we studied effects chromatin condensation histone acetylation levels male elegans. We found that H4K16 acetyltransferase MYS-1/Tip60 mediates dramatic decondensation as measured by FISH. However, RNA-seq analysis revealed MYS-1 contributes only slightly chromosome. These results suggest level does necessarily correlate with degree change vivo. Furthermore, sensitive MYS-1-mediated than autosomes, despite similar H4K16ac all chromosomes, ChIP-seq. weakly highly expressed genes do contain exceptionally high H4K16ac. indicate influence regulation chromosome; however, they fully account for observed
