摘要: Abstract Human umbilical vein endothelial cells differentiate within 12 h to form capillary-like networks of tube structures when the are plated on Matrigel, a mixture basement membrane proteins. Nothing is known about intracellular signaling events involved in this differentiation. As first step define process, we investigated possible role protein kinase C activation by β-phorbol 12-myristate 13-acetate (PMA) regulating formation structures. In model, PMA increased several-fold dose-dependent manner with half-maximum stimulation at approximately 5 nM PMA. absence serum, essentially little or no tubes were formed Matrigel unless was added medium. Only active phorbol analogs formation, while inhibitor, H-7, blocked formation. The activators and inhibitors effective only just after plating did not affect already tubes. This study suggests that early vitro cell Matrigel.
