Andrographolide ameliorates diabetic nephropathy by attenuating hyperglycemia-mediated renal oxidative stress and inflammation via Akt/NF-κB pathway

摘要: Diabetic nephropathy (DN) is characterized by proliferation of mesangial cells, hypertrophy and extracellular matrix (ECM) accumulation. Our recent study found that andrographolide inhibited high glucose-induced cell fibronectin expression through inhibition AP-1 pathway. However, whether has reno-protective roles in DN not been fully elucidated. Here, we studied the pharmacological effects against progression dysfunction. Diabetes was induced C57BL/6 mice intraperitoneal injection streptozotocin (STZ). After 1 weeks after STZ injection, normal diet substituted with a high-fat (HFD). were injected (2 mg/kg, twice week). 8 weeks, functional histological analyses carried out. Parallel experiments uncovering molecular mechanism which prevents from performed cells. Andrographolide increases fasting blood glucose, triglyceride, kidney/body weight ratio, urea nitrogen, serum creatinine 24-h albuminuria diabetic mice. also prevented renal ECM Furthermore, markedly attenuated NOX1 expression, ROS production pro-inflammatory cytokines as well. Additionally, Akt/NF-κB signaling These results demonstrate protective experimental inhibiting oxidative stress, inflammation fibrosis.
