Berberine attenuates high glucose-induced proliferation and extracellular matrix accumulation in mesangial cells: Involvement of suppression of cell cycle progression and NF-κB/AP-1 pathways

摘要: Berberine has been shown to have renoprotective effects on diabetes through attenuating TGF-β1 and fibronectin (FN) expression. However, how berberine regulates FN is not fully clear. Here we investigated whether inhibited expression in high glucose-cultured mesangial cells. significantly cell proliferation hypertrophy by increasing the population G1-phase reducing that S-phase. In addition, reversed glucose-induced down-regulation of cyclin-dependent kinase inhibitor p21Waf1/Cip1 p27Kip1. p65 translocation nucleus c-jun phosphorylation induced glucose. Furthermore, attenuated FN. Using a luciferase reporter assay, found transcription activity NF-κB AP-1 was blocked berberine. Electrophoretic mobility shift assay showed glucose increased DNA binding activity. These data indicate modulating cycle progress. suppressed blocking NF-κB/AP-1 pathways.
