Utility of Polymerase Chain Reaction for Analysis of Antigen Receptor Rearrangement in Staging and Predicting Prognosis in Dogs with Lymphoma

摘要: In lymphoid neoplasia, molecular assays to confirm clonality rely on the fact that cells normally contain DNA regions with unique sequences, resulting from recombination of V, D, and J genes. The purpose this study was determine utility polymerase chain reaction (PCR) for antigen receptor rearrangements (PARR) staging predicting prognosis in canine lymphoma. We hypothesized PARR assay would offer a sensitive method detecting neoplastic blood, presence such be negative prognostic finding compared dogs no detectable circulating tumor cells. Eighty-six patients histologically or cytologically confirmed lymphoma were studied initial diagnosis until death euthanasia. All had representative tumor-infiltrated lymph node peripheral whole blood. Sixty-two clonal detected able staged by PARR. Seventeen (27%) their blood 45 (73%) positive. Our findings showed (1) correlated clinical stage more likely detect 5 lymphomas, (2) than visual assessment bone marrow because 80% 3 lymphomas blood-PARR-positive, (3) PCR not disease-free interval (DFI) survival.
