摘要: O6-methylguanine-DNA methyltransferase (MGMT) mRNA expressions were examined in 100 neuroepithelial tumors by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR) using SYBR Green I. The mean relative quantitation value of MGMTmRNA normalized to the level β2-microglobulin for was 5.3 ± 11.2. 41 glioblastomas significantly higher than that other 59 (p = 0.0008 Student's t-test). In contrast, means 19 low-grade gliomas and 12 medulloblastomas lower 0.0282 p 0.0456 Among 55 retrospective patients who had been treated with 1-(4-amino-2-methyl-5-pyrimidynyl)methyl-3-(2-chloroethyl)- 3-nitrosourea hydrochloride (ACNU), a significant independent predictor effect initial therapy ACNU 0.0007 Mann-Whitney U-test) survival period 0.0175 Wald test). ≧1 most factor predicting treatment multi-variant regression analysis < 0.0001). These results suggest our individual adjuvant based on expression may be improved application RT-PCR. © 2002 Wiley-Liss, Inc.
